Most Serious Warnings
Estradiol should not be used to prevent heart disease or dementia. Studies of estrogen, with or without progestin, have shown increased risks of:
Heart attack
Stroke
Blood clots
Breast cancer
Dementia
Use of estrogen alone in women with a uterus increases the risk of endometrial cancer. A progestin may be prescribed to lower this risk. Estradiol should be used at the lowest effective dose for the shortest duration consistent with treatment goals
Who Should NOT Take Estradiol
Do not take this medication if you:
Have a known allergy to estradiol or any ingredient in the formulation
Have or have had blood clots, stroke, or heart attack
Have known or suspected breast cancer or estrogen-dependent tumors
Have liver disease or liver tumors
Have unexplained vaginal bleeding
Have thickening of the uterine lining (endometrial hyperplasia) without evaluation
Are pregnant or may become pregnant
Before Taking Estradiol, Tell Your Healthcare Provider if You:
Have high blood pressure, diabetes, or high triglycerides
Have a history of blood clots or a family history of clotting disorders
Smoke or use tobacco products
Have gallbladder disease, liver problems, or migraines
Have depression or memory concerns
Will be immobilized for long periods such as after surgery or during travel
Take prescription, over the counter, or herbal medications
Possible Side Effects
Common (mild to moderate):
Nausea or upset stomach
Breast tenderness
Headache
Bloating or fluid retention
Vaginal discharge or irregular bleeding
Skin darkening on the face
Serious (seek medical care immediately):
Chest pain, shortness of breath, coughing up blood
Swelling, warmth, redness, or pain in an arm or leg
Sudden severe headache, confusion, weakness on one side of the body, or vision changes
Yellowing of the skin or eyes, dark urine, or severe abdominal pain
New breast lump or nipple discharge
Unusual vaginal bleeding
Signs of a severe allergic reaction such as swelling of the face, lips, tongue, or throat
While Taking Estradiol
Tell your healthcare provider immediately if you:
Develop symptoms of a blood clot or stroke
Notice abnormal vaginal bleeding
Experience vision changes or severe headaches
Develop breast changes or new lumps
Regular breast exams, pelvic exams, and monitoring of blood pressure, labs, and bone density may be recommended
Pregnancy and Fertility
Estradiol should not be used during pregnancy. If you become pregnant while taking this medication, contact your provider immediately
Overdose
Symptoms of overdose may include nausea, vomiting, breast tenderness, dizziness, abdominal pain, or vaginal bleeding. If overdose is suspected, seek emergency medical care or contact Poison Control immediately.
Missed Dosage
If you miss a dose, take it as soon as you remember unless it is almost time for your next dose. Do not double your dose.

Store at room temperature as directed on your pharmacy label
Keep out of reach of children and pets
Dispose of unused or expired medication according to local regulations
